A hands-on-workshop on Ten-fold Robust Expansion Microscopy (TREx)
Ten-fold Robust Expansion Microscopy (TREx) is a super-resolution imaging technique that enables nanoscale
visualization of proteins using conventional fluorescence microscopes. By physically expanding biological
samples, TREx increases the spatial separation between fluorophores by approximately tenfold, allowing
researchers to resolve fine cellular structures beyond the diffraction limit. This hands-on workshop provides
comprehensive training in applying TREx to cultured human cell lines. Participants will gain practical experience
in the complete experimental workflow, including:

Sample preparation and fixation

Immunofluorescence staining of proteins of interest

Sample expansion using the TREx protocol

High-resolution imaging of expanded samples using the Zeiss Airyscan microscope

Best practices for data acquisition and troubleshooting​
Target audience

PhD students and early-career postdoctoral researchers whose group leader is a member of the Center for Synthetic Genomics (SynGen).
Prerequisites

Previous experience working in an S1 laboratory is mandatory.

Familiarity with basic cell culture and cytological techniques is required.

Course Format
The workshop will be held in person at KIT. Course hours are 09:30-16:30 (1 long day till 19:00)
Additional Notes
Due to the hands-on nature of the training, attendance in person is mandatory.
For participants traveling from Mainz, accommodation during the course may be arranged upon request, subject to availability.
Selection Criteria
As the workshop is limited to 8 participants, applicants are asked to briefly describe in the registration form how
their research will benefit from participation in this workshop. Applications will be carefully reviewed based on
the submitted motivation statement. Following the evaluation process, applicants will be informed of the
outcome by 14 September 2026.
